  1. General Information

    1. The Leica DM LB is the premiere optical microscope in the BYU Cleanroom. Its high resolution, precision imaging allows its users to get an accurate, detailed view of their work. The Leica is also equipped with a video camera and computer linkup which makes capturing and sharing the microscopes images simple.
  2. Microscope Lens Calibration

    1. LENS_________DISTANCE BETWEEN NUMBER MARKS
    2. X10______________________123 microns
    3. X20_______________________60 microns
    4. X50_______________________24 microns
    5. X100______________________12 microns
  3. Function Instructions

    1. Power, Brightness, Focus
    2. Stage Movement
    3. Aperture Diaphragm
      1. The aperture diaphragm can be narrowed to decrease the amount of light hitting the specimen. This will decrease reflections off the specimen and increase the image contrast. However, closing the aperture too much will cause a loss in detail.
    4. Field Diaphragm
      1. The field diaphragm can be narrowed to decrease the field of view.
    5. Polarizer
      1. Using the polarizer allows the user of the microscope to polarize the light before it reaches the sample. This is done by pushing in the polarizing lens on the right side of the microscope. The analyzer will show how the light was affected by reflecting off the sample. First, push in the analyzer than rotate the lens to see the how the light polarization has changed.
    6. Capturing Images on the Computer
      1. Turn on the camera by flipping the switch on the black box behind the microscope. (the green light will turn on on top of the camera)
      2. Click on the FBG32 icon on the computer desktop.
      3. In the program the menu tab furthest right will read either GRAB or LIVE.
        1. If it says GRAB the program is showing the live feed from the camera and the user has the option of grabbing the current image.
        2. It it says LIVE, the program is showing the last image grabbed and the user can press live to see the live feed from the camera.
      4. Save option is under the file tab.
